چکیده
The delta-amino acid 5-aminolevulinic (ALA), is the precursor of endogenous photosensitiser Protoporphyrin IX (PpIX), and currently approved for Photodynamic Therapy (PDT) certain superficial cancers. However, ALA-PDT not very effective in diseases which T-cells play a significant role. Cutaneous T-cell lymphomas (CTCL) group non-Hodgkin malignant diseases, includes mycosis fungoides (MF) Sézary syndrome (SS). In previous work, we have designed new ALA esters synthesised by three-component Passerini reactions, some them showed higher performance as compared to ALA. This work aimed determine efficacy pro-photosensitisers five 2-hydroxy-N-arylacetamides (1f, 1 g, h, 1i k) lipophilicity than Myla cells MF HuT-78 SS. We also tested its effectiveness against already marketed methyl ester (Me-ALA) hexyl (He-ALA). Both cell SS were effectively equally photoinactivated ALA-PDT. Besides, concentration required induce half maximal porphyrin synthesis was 209 μM 169 cells. As criterion efficacy, calculated derivatives necessary plateau values obtained from These achieved at concentrations 4 12 times lower ALA, according derivative used. For He-ALA, 24 25 inducing comparable both CTCL light doses 50% death (LD50) 1f, h around 18 J/cm2 respectively, after exposure 0.05 mM compounds. On other hand, LD50s compound k 40 57 HuT-78, respectively. contrast, Me-ALA did provoke photokilling since employed far below saturation point these Our results suggest potential use topical application PDT treatment extracorporeal depletion activated

Derivatives of 5-Aminolevulinic Acid for Photodynamic Therapy
Photodynamic therapy (PDT) is a clinical treatment that combines the effects of visible light irradiation with subsequent biochemical events that arise from the presence of a photosensitising drug (possessing no dark toxicity) to cause destruction of selected cells. Today, the most common agent used in dermatological PDT is 5-aminolevulinic acid (ALA). As a result of its hydrophilic character, ...

Background: Basal Cell Carcinoma (BCC) is the most common type of skin cancer in human beings. Photodynamic therapy (PDT) is a novel therapeutic method which may be regarded as a non-invasive useful alternative for traditional treatments of BCC. This study was designed with the aim of evaluating the primary response of BCC to PDT.Methods: This clinical trial was perform...
متن کامل[Apoptosis-inducing effect of 5-aminolevulinic acid-mediated photodynamic therapy (5-ALA-PDT) on cervical cancer cell lines].
BACKGROUND & OBJECTIVE 5-Aminolevulinic acid-mediated photodynamic therapy (ALA-PDT) is a new ablation treatment for tumors, while its function mechanism in cervical cancer is unclear. This study was to investigate the effects of ALA-PDT on cervical cancer cell lines. METHODS The effects of ALA-PDT on proliferation of 8 human cervical cancer cell lines were examined by MTT assay to find out t...
متن کاملAminolevulinic Acid-Mediated Photodynamic Therapy of Human Meningioma: An in Vitro Study on Primary Cell Lines
OBJECTIVE Five-aminolevulinic acid (5-ALA)-induced porphyrins in malignant gliomas are potent photosensitizers. Promising results of ALA-PDT (photodynamic therapy) in recurrent glioblastomas have been published. Recently, 5-ALA-induced fluorescence was studied in meningioma surgery. Here, we present an experimental study of ALA-PDT in an in vitro model of primary meningioma cell lines. METHOD...
